(AP) ? Russian's desire to keep its last remaining ally in the Middle East collided head-long with the United States' desire to remove President Bashar Assad and replace him with a democracy at a pivotal United Nations-brokered conference Saturday seeking a political solution to the violence in Syria.

Efforts at bridging the Russian-U.S. divide over Syria held the key to international envoy Kofi Annan's plan for easing power away from Assad's grip and ending 16 months of horrific violence in Syria before it erupts into full-blown civil war.

Without agreement among the major powers on how to form a transitional government for Syria, Assad's regime which is Iran's closest ally would be emboldened to try to remain in power indefinitely ? and that would also complicate the U.S. aim of preventing Iran from getting a nuclear weapon.

At talks Friday night, top U.S. and Russian diplomats remained deadlocked over the negotiating text to agree on guidelines and principles for "a Syria-led transition."

Hopes have centered on persuading Russia, which is Syria's most important ally, protector and supplier of arms, to agree to a plan that would end the Assad family dynasty that has ruled Syria for more than four decades. But the Russians want Syria alone to be the master of its fate, at a time when Assad's regime and the opposition are increasingly bitterly polarized.

"Ultimately, we want to stop the bloodshed in Syria. If that comes through political dialogue, we are willing to do that," said Khalid Saleh, a spokesman for the Syrian National Council, a coalition of Syrian opposition groups in Istanbul, Turkey. "We are not willing to negotiate (with) Mr. Assad and those who have murdered Syrians. We are not going to negotiate unless they leave Syria."

The negotiating text for the multinational conference calls for establishing a transitional government of national unity, with full executive powers, that could include members of Assad's government and the opposition and other groups. It would oversee the drafting of a new constitution and elections.

But the text that would serve as the framework for Annan's peace efforts also would "exclude from government those whose continued presence and participation would undermine the credibility of the transition and jeopardize stability and reconciliation."

Foreign ministers from all five of the permanent, veto-wielding members of the U.N. Security Council ? Britain, China, France, Russia and the U.S. ? converged at the U.N.'s European headquarters in the sprawling Palais des Nations overlooking Lake Geneva and Mont Blanc. Russia and China have twice use their council veto to shield Syria from U.N. sanctions.

For his "Action Group on Syria," U.N.-Arab League envoy Annan also invited Turkey, the U.N. secretary-general Ban Ki-moon, the European Union and Iraq, Kuwait and Qatar as heads of three groups within the League of Arab States.

Major regional players Iran and Saudi Arabia were not invited. The Russians objected to the Saudis, who support the Syrian opposition. The U.S. objected to the Islamic Republic, which supports Assad's regime.

Much of the work remains to be hammered out by Russian Foreign Minister Sergey Lavrov and U.S. Secretary of State Hillary Rodham Clinton, who met for an hour in St. Petersburg before sharing dinner Friday before Clinton left Russia. Lavrov predicted the meeting had a "good chance" of finding a way forward, despite the grim conditions on the ground.

Russia insists that outsiders cannot order a political solution for Syria, while the U.S. is adamant that Assad should not be allowed to remain in power at the top of the transitional government.

But Clinton said Thursday in Riga, Latvia, that all participants in the Geneva meeting, including Russia, were on board with the transition plan. She told reporters that the invitations made clear that representatives "were coming on the basis of (Annan's) transition plan."

The uprising in Syria since March of last year has killed some 14,000 people and Syria has shot down a Turkish warplane. Turkey has responded by setting up anti-aircraft guns on its border with Syria. On Friday, Syrian troops shelled a suburb of Damascus, killing an estimated 125 civilians and 60 soldiers.

The United Nations says the violence in Syria has worsened since a cease-fire deal in April and the bloodshed appears to be taking on dangerous sectarian overtones, with growing numbers of Syrians targeted on account of their religion. The increasing militarization of both sides in the conflict has Syria heading toward civil war.

Programmable DNA scissors found for bacterial immune system

ScienceDaily (June 28, 2012) ? Genetic engineers and genomics researchers should welcome the news from the Lawrence Berkeley National Laboratory (Berkeley Lab) where an international team of scientists has discovered a new and possibly more effective means of editing genomes. This discovery holds potentially big implications for advanced biofuels and therapeutic drugs, as genetically modified microorganisms, such as bacteria and fungi, are expected to play a key role in the green chemistry production of these and other valuable chemical products.

Jennifer Doudna, a biochemist with Berkeley Lab's Physical Biosciences Division and professor at the University of California (UC) Berkeley, helped lead the team that identified a double-RNA structure responsible for directing a bacterial protein to cleave foreign DNA at specific nucleotide sequences. Furthermore, the research team found that it is possible to program the protein with a single RNA to enable cleavage of essentially any DNA sequence.

"We've discovered the mechanism behind the RNA-guided cleavage of double-stranded DNA that is central to the bacterial acquired immunity system," says Doudna, who holds appointments with UC Berkeley's Department of Molecular and Cell Biology and Department of Chemistry, and is an investigator with the Howard Hughes Medical Institute (HHMI). "Our results could provide genetic engineers with a new and promising alternative to artificial enzymes for gene targeting and genome editing in bacteria and other cell types."

Doudna is one of two corresponding authors of a paper in the journal Science describing this work titled "A programmable dual RNA-guided DNA endonuclease in adaptive bacterial immunity." The second corresponding author is Emmanuelle Charpentier of the Laboratory for Molecular Infection Medicine at Sweden's Ume? University. Other co-authors of the paper were Martin Jinek, Krzysztof Chylinski, Ines Fonfara and Michael Hauer.

Bacterial and archaeon microbes face a never-ending onslaught from viruses and invading circles of nucleic acid known as plasmids. To survive, the microbes deploy an adaptive-type nucleic acid-based immune system that revolves around a genetic element known as CRISPR, which stands for Clustered Regularly Interspaced Short Palindromic Repeats. Through the combination of CRISPRs and associated endonucleases, called CRISPR-associated -- "Cas" -- proteins, bacteria and archaeons are able to utilize small customized crRNA molecules (for CRISPR-derived RNA) to target and destroy the DNA of invading viruses and plasmids.

There are three distinct types of CRISPR/Cas immunity systems. Doudna and her colleagues studied the Type II system which relies exclusively upon one family of endonucleases for the targeting and cleaving of foreign DNA, the Cas9 proteins.

"For the Type II CRISPR/Cas system, we found that crRNA connects via base-pairs with a trans-activating RNA (tracrRNA), to form a two-RNA structure," Doudna says. "These dual RNA molecules (tracrRNA:crRNA) direct Cas9 proteins to introduce double-stranded DNA breaks at specific sites targeted by the crRNA-guide sequence."

Doudna and her colleagues demonstrated that the dual tracrRNA:crRNA molecules can be engineered as a single RNA chimera for site-specific DNA cleavage, opening the door to RNA-programmable genome editing.

"Cas9 binds to the tracrRNA:crRNA complex which in turn directs it to a specific DNA sequence through base-pairing between the crRNA and the target DNA," Doudna says. "Microbes use this elegant mechanism to cleave and destroy viruses and plasmids, but for genome editing, the system could be used to introduce targeted DNA changes into the genome.

Doudna notes that the "beauty of CRISPR loci" is that they can be moved around on plasmids.

"It is well-established that CRISPR systems can be transplanted into heterologous bacterial strains," she says. "Also, there is evidence to suggest that CRISPR loci are horizontally transferred in nature."

Doudna and her colleagues are now in the process of gathering more details on how the RNA-guided cleavage reaction works and testing whether the system will work in eukaryotic organisms including fungi, worms, plants and human cells.

"Although we've not yet demonstrated genome editing, given the mechanism we describe it is now a very real possibility," Doudna says.

Princess Merida (voiced by Kelly Macdonald) in Pixar?s ?Brave? shows girls that they can shoot a bow and arrow, too.

It may be clearer than ever that Pixar?s golden era went out with ?Toy Story 3,? the jewel in the crown of an animation studio that ruled over its medium for 15 glorious years. But does ?Brave,? Pixar?s new badass-Scottish-lass adventure, deserve to be flogged simply because it?s not ?WALL?E?-level transcendent? Yes and no. Solid yet innocuous, moving yet minimally ambitious, this old-fashioned fantasy is an oxymoronic specimen, aiming for progressiveness while clinging to safe traditions. But it nevertheless offers something rarely seen in the world of toons: A mother-daughter tale that leaves the boys on the side.

Merida (voiced by Kelly Macdonald) is your typical oppressed tomboy princess, a nervy athlete who?d rather fire arrows than follow the rules of her mother (voiced by Emma Thompson), a stern queen trying to marry Merida off. Before you can say ?Mulan,? our free-thinking heroine is pulling an Ariel, consulting a shady witch to ?change her fate.? What results is as zanily random as it is intentionally gender-subverting, and when not inducing yawns with lazy plot offshoots (the male characters simply holler and spar), the combo yields tender common ground for its feuding kinswomen.

Directed by Pixar?s Mark Andrews and Brenda Chapman, ?Brave? follows Disney?s ?Tangled? as a film in which hair proves a benchmark in animation?s evolution (Merida?s springy, scarlet locks are wondrous to behold). But, more importantly, the movie boasts Pixar?s first female lead, a separate benchmark many will view as a blown opportunity.

Andrews and Chapman may pilot their film to an affecting resolution, but their introductory scene of Merida triumphantly scaling a mountain (one of many moments with misty vistas), is hardly a promise that?s kept. ?Brave? is an atmospheric jaunt with quirky flourishes, but its bound to leave the gals in the audience feeling a bit shortchanged. Whereas, say, ?Up? presented a man with startling human complexity, Pixar?s unwittingly regressive 13th feature keeps things light and unchallenging, all while quietly waving a flag of feminism. Instead of passing women the ball, it merely throws them a bone.

Willem Dafoe fans shouldn?t miss the eco-drama ?The Hunter,? which sees the chameleonic actor take the lead as a sharp-shooting mercenary, tasked to hunt down an endangered animal in Tasmania. Shot on location, the film is predominantly picturesque, and it ably blends environmental themes with taut conflict. Its scattered missteps are overshadowed by Dafoe, who gives a soulful performance as the noirish antihero.

    Mom & Dad's 40th Anniversary SURPRISE Party!

    One year ago (July 1st), on my parents' 39th wedding anniversary, I called my brother and told him that we had 1 year to plan mom & dad's 40th anniversary SURPRISE party.? He was on board and we quickly started throwing ideas around.? It wasn't until?January that we really started to get the ball rolling, as far as planning goes.
    My uncle Steve (mom's brother) was kind enough to allow us to use his gorgeous home.? We also hired "The Taco Man".? There were many details to plan and execute and I can now say that everything turned out PERFECT!? The best part..... THEY WERE ACTUALLY SURPRISED!? I am still shocked that no one blew it.? I didn't think that ever happened anymore.? It seems like someone always spills the beans.?
    Here is how it worked:? Cade's birthday is in a few weeks, so I told my parents that we were having a "family only" party at uncle Steve's house for Cade.? I told them that we had to have it a few weeks before his bday since some of Brad's family would be out of town and this was the only weekend that worked.? I wanted my parents to arrive at 1pm, so I told them Cade's bday party started at 2pm, and could they please arrive at 1pm to help Brad & I set up.? They said, "Of course we will help!"? (But really, everyone was arriving at 12:30pm.? So this would give everyone time to arrive before mom & dad arrived at 1pm.)

    We invited our huge family and also 5 couples that are my parents very best friends.? My parents were SO happy that all of their closest friends were there!? Here?are a bunch of them.

    I am SO happy that mom & dad were surprised!? I will post a video soon that shows them walking in and hearing "SURPRISE!".? You can tell by my mom's face that they had no clue.? It was really neat how we let them know that my brother and family had flown in from Texas.? Basically, they were in a back bedroom hiding.? Once we all said, "Surprise!", and our hearts stopped racing, ?my mom said, "Oh my gosh!? Everyone's here.? Is Chris here too?"? I said, "No, but he wanted me to call him so that he could say hi and hear your reaction."? So, I called him right there and my mom was talknig to him and telling him how neat this was, etc.? She then gave the phone to my dad so that he could talk to Chris.? About 2 minutes later, Chris walks out of the room still on the phone!? I thought my parents were going to have a heart attack.? It was SO NEAT!? I will post the video soon!?

    Exxon's CEO: Climate, energy fears overblown

    ExxonMobil CEO Rex Tillerson says fears about climate change, drilling, and energy dependence are overblown.

    In a speech Wednesday, Tillerson acknowledged that burning of fossil fuels is warming the planet, but said society will be able to adapt. The risks of oil and gas drilling are well understood and can be mitigated, he said. And dependence on other nations for oil is not a concern as long as access to supply is certain, he said.

    Tillerson blamed a public that is "illiterate" in science and math, a "lazy" press, and advocacy groups that "manufacture fear" for energy misconceptions in a speech at the Council on Foreign Relations.

    He highlighted that huge discoveries of oil and gas in North America have reversed a 20-year decline in U.S. oil production in recent years. He also trumpeted the global oil industry's ability to deliver fuels during a two-year period of dramatic uncertainty in the Middle East, the world's most important oil and gas-producing region.

    "No one, anywhere, any place in the world has not been able to get crude oil to fuel their economies," he said.

    In his speech and during a question-and-answer session after, he addressed three major energy issues: Climate change, oil and gas drilling pollution, and energy dependence.

    Tillerson, in a break with predecessor Lee Raymond, has acknowledged that global temperatures are rising. "Clearly there is going to be an impact," he said Wednesday.

    But he questioned the ability of climate models to predict the magnitude of the impact. He said that people would be able to adapt to rising sea levels and changing climates that may force agricultural production to shift.

    "We have spent our entire existence adapting. We'll adapt," he said. "It's an engineering problem and there will be an engineering solution."

    Andrew Weaver, chairman of climate modeling and analysis at the University of Victoria in Canada, disagreed with Tillerson's characterization of climate modeling. He said modeling can give a very good sense of the type of climate changes that are likely. And he said adapting to those changes will be much more difficult and disruptive than Tillerson seems to be acknowledging.

    Steve Coll, author of the recent book "Private Empire: ExxonMobil and American Power," said he was surprised Exxon would already be talking about ways society could adapt to climate change when there is still time to try to avoid its worst effects. Also, he said, research suggests that adapting to climate change could be far more expensive than reducing emissions now. "Moving entire cities would be very expensive," he said.

    Legislation or regulation that would help slow the emissions of global warming gases would likely lead to lower demand for oil and gasoline, and could reduce Exxon's profit.

    Tillerson expressed frustration at the level of public concern over new drilling techniques that tap natural gas and oil in shale formations under several states. He said environmental advocacy groups that "manufacture fear" have alarmed a public that doesn't understand drilling practices ? or math, science or engineering in general. He blamed "lazy" journalists for producing stories that scare the public but don't investigate the claims of advocacy groups.

    Drilling for oil and gas will always involve risks of spills and accidents, he said. But those risks are manageable and worth taking because they are small given the amount of energy they produce.

    Drilling in shale formations, he said, only poses a small risk to those living nearby. It is neither life threatening nor long lasting and can be controlled in the event of an accident.

    Drillers force millions of gallons of water mixed with sand and some hazardous chemicals into shale formations. The technique breaks up rock and creates escape routes for oil and gas. If the drilling wastewater is not treated properly or if it seeps through cracked drilling pipes, it could contaminate drinking water.

    The industry's biggest challenge, he said, is "taking an illiterate public and try to help them understand why we can manage these risks."

    Tillerson made a distinction between energy security and energy dependence. He said that energy security ? making sure that the economy has access to energy ? is crucial.

    But he said access to energy is not in peril. "Some of the fears around energy security are not well founded," he said.

    The quest for energy independence, though, is misguided, he said. It doesn't matter where the U.S. gets oil because crude is priced globally. Even if the U.S. used only oil from North America, a disruption in the Middle East would increase global prices, hurt the U.S. and global economies, and force Americans to pay more at the pump.

    Even if the U.S. no longer needed Middle Eastern oil, it would likely want to play a major role in helping maintain the region's security, Tillerson said.?

    Jiayuh Lin, PhD, and colleagues at Nationwide Children's Hospital have developed a drug to target the most common cancerous bone tumor in children, osteosarcoma, using a version of the FDA-approved drug, Celebrex. The team will soon begin testing the drug using human and canine tumor cell lines thanks to a two-year, $200,000 grant from Alex's Lemonade Stand Foundation for Childhood Cancer.

    Osteosarcoma is an aggressive bone tumor that usually develops during the period of rapid growth that occurs in adolescence. A signaling pathway known as the STAT3 pathway is common in osteosarcoma and is crucial to tumor formation and cancer progression. Blocking STAT3 signaling is considered a potential approach for treating osteosarcoma; however, few drugs are available that can inhibit STAT3 and be clinically relevant.

    "One of the main barriers to developing a clinical drug to inhibit STAT3 is finding lead compounds that exhibit desirable drug properties," says Dr. Lin, who is a principal investigator in the Center for Childhood Cancer at The Research Institute at Nationwide Children's Hospital.

    Dr. Lin and associate professor at The Ohio State University College of Pharmacy, Chenglong Li, PhD, have found that developing STAT3-selective inhibitors can be accelerated by pairing a novel STAT3 drug discovery method with drug repositioning techniques to create inhibitors with desirable drug properties. Using this method, Dr. Lin's team has developed a STAT3-selective inhibitor, 8A, using the FDA-approved drug, celecoxib (brand name: Celebrex, Pfizer, New York ). Celecoxib is typically prescribed to relieve pain, tenderness, swelling and symptoms of inflammatory conditions like osteoarthritis. 8A is more potent and selective than celecoxib against STAT3 signaling in osteosarcoma cells.

    The new funding will allow Dr. Lin's team to develop two additional 8A analogs that would further increase STAT3 binding, while retaining the drug properties of celecoxib. The team will then test all three versions in human and canine osteosarcoma cell lines and in a mouse tumor model. Future research will include collaboration with Cheryl London, DVM, PhD, at The Ohio State University College of Veterinary Medicine to initiate clinical trials of a lead 8A analog in dogs with spontaneous osteosarcoma. Osteosarcoma in dogs is very similar to osteosarcoma in humans. Trials will provide valuable data for human trials as well as help determine the feasibility of using lead 8A analog to treat these tumors in canines.

    "This is a unique approach in osteosarcoma targeted therapy," says Dr. Lin. "We feel confident that our findings will advance the field of childhood osteosarcoma treatments."

    ?The Ultimate Fighter? makes changes, but will it be enough?

    Usually, in the months before the UFC starts taping a season of "The Ultimate Fighter," the show holds open tryouts. Fighters from all over the country flock to hotel ballrooms to try to win the favor of the UFC execs watching their every move.

    As the show heads into its 16th season, there were no tryouts. Instead, the UFC asked managers to bring welterweights to Las Vegas for auditions and personal interviews.

    "I've never seen them skip the tryout process and fly guys straight in," one manager told MMAjunkie.com.

    This isn't the only change coming to the UFC's long-running reality show. As Kevin Iole reported, the UFC is going back to a taped format after the 15th season aired fights live. FX has also decided to keep the show on Friday nights.

    Though ratings are up for FX on Friday nights, the ratings for the show are down compared to previous seasons. Attendance and gate from the show's finale were also down.

    TUF is first a television show, and two rules have always remained true about the modern television landscape. First, it's hard to keep any show fresh for 16 seasons. Secondly, Friday nights are where ratings go to die. There is not much the UFC can do to get around those truisms.

    Mom and baby carjacked outside Starbucks

    Dallas police are searching for a man who carjacked a woman and her baby Sunday afternoon after stealing from a nearby Walmart.

    Police said the man stole a t-shirt from the store and then jumped into the woman's car in a Starbucks parking lot while police were chasing him.

    The man didn't realize the woman and her 15-month-old daughter were sitting inside the vehicle, police said.


    "I thought we were dead," Kari Kimbler said. "I thought he was going to take us somewhere and dump us off or kill us. I had no idea what was going to happen."

    Kimbler said her husband had gone inside the Starbucks when the man jumped in the driver's seat and took off. She said he started driving erratically, going the wrong way down Cockrell Hill Road.

    "I started hitting him and beating him and screaming, ?My baby, my baby -- I have my baby. Please let us out. Please dump us out. I don't care where it is. Please let us out,'" Kimbler said.

    She said the man initially refused.

    "Then momma bear kicked, and I all could think was, 'I've got to protect my baby with all that I have,' and it came out, and I just beat the living daylights out of him as hard as I could -- in his head, just punching in his head, and she was screaming too," Kimbler said.

    Kimbler said the man stopped underneath a bridge after five minutes.

    "He did pull over," she said. "But as I was getting her out, he pushed us out to get us out and just kept going."

    Kimbler said she can breathe easier now that she and her baby are home and safe. But she said the fear of what could have happened is still with her.

    "I've told all of my loved ones how much I love them," she said. "And I hug my daughter tighter, and I told her tighter, and I tell her how much mommy and daddy love her."

    "I'm just so thankful she and I got out alive," Kimbler said.

    Dallas police described the man as Latino, approximately 5-foot-9-inches tall, in his early 30s, who weighs about 185 pounds with a bald head and sleeves of tattoos on both arms. He was last seen wearing a gray Texas Longhorns hat, khaki shorts and a dark-colored shirt.

    Bobby Brown and Alicia Etheridge: Married!


    Bobby Brown and fiancée Alicia Etheridge got married Monday.

    Bobbi Kristina Brown? Not on hand for the Hawaiian celebration.

    The R&B star and former husband of Whitney Houston was surrounded by other family and friends, including his older kids Landon, 23, La'Princia, 22, and Bobby Jr., 19, as well as Brown and Alicia Etheredge's only child together, Cassius, 3.

    The groom wore a red suit and Adidas sneakers ... obviously.

    Bobby, who has been arrested numerous times over the years for suspicion of DUI, not paying child support and more, posted the photo of the wedding party.

    The couple became engaged in a very public way in 2010, when Brown popped the question to Etheridge during a New Edition show in Jacksonville, Fla.

    Brown has often credited Etheredge with giving him a new lease on life following his 2007 divorce from Whitney Houston, who passed away in February.

    Bobbi Kristina Brown, 19, Bobby's daughter with Whitney Houston, is "not on good terms right now" with her dad and was MIA, according to a source.

    As for a honeymoon ... Brown and Etheridge, who is also his manager, will continue traveling with the New Edition tour, resuming June 23 in Oakland, Calif.

    One afternoon last August, at a hospital on the outskirts of Los Angeles, a former beauty queen named Emma Coronel gave birth to a pair of heiresses. The twins, who were delivered at 3:50 and 3:51, respectively, stand to inherit some share of a fortune that Forbes estimates is worth a billion dollars. Coronel's husband, who was not present for the birth, is a legendary tycoon who overcame a penurious rural childhood to establish a wildly successful multinational business. If Coronel elected to leave the entry for "Father" on the birth certificates blank, it was not because of any dispute over patrimony. More likely, she was just skittish about the fact that her husband, Joaqu?n Guzm?n, is the C.E.O. of Mexico's Sinaloa cartel, a man the Treasury Department recently described as the world's most powerful drug trafficker. Guzm?n's organization is responsible for as much as half of the illegal narcotics imported into the United States from Mexico each year; he may well be the most-wanted criminal in this post-Bin Laden world. But his bride is a U.S. citizen with no charges against her. So authorities could only watch as she bundled up her daughters and slipped back across the border to introduce them to their dad.
